Total RNA containing small RNA was extracted from mouse kidney by using the Trizol reagent (Invitrogen) and purified with mirVana miRNA Isolation Kit (Ambion, Austin, TX, USA) according to manufacter’s protocol. The purity and concentration of RNA were determined from OD260/280 readings using spectrophotometer (NanoDrop ND-1000). RNA integrity was determined by 1% formaldehyde denaturing gel electrophoresis.

100 ng purified RNA was dephosphorylated and ligated with pCp-Cy3 using the Agilent miRNA labeling reagent according to the manufacturer's instructions.

The labeled miRNA was hybridized to the Agilent miRNA microarray designed with eight identical arrays per slide (8 × 60K format), with each array containing probes interrogating 1247 mouse miRNAs from the Sanger database (Version 19.0) according to the manufacturer’s protocol

Images were scanned with the Agilent microarray scanner (Agilent), gridded, and analyzed using Agilent feature extraction software version 10.10.
