Treatment protocol |
After 6 days, 3x106 DCs, obtained from three different healthy donors, were transfected with either miR-29b or NC and co-cultured with U266 MM cells. 24 hours after transfection, cell were harvested and separated with immuno-magnetic microbeads.

Growth protocol |
DCs were generated from healthy donors' CD14+ monocytes isolated by immunomagnetic separation. These cells were then cultured for 6 days in RPMI-1640 medium containing 10% FBS, 1% penicillin/streptomycin, 100 ng/ml recombinant human GM-CSF and 50 ng/ml IL-4 . Half-medium was replaced every 48 hours.
