Treatment protocol After 6 days, 3x106 DCs, obtained from three different healthy donors, were transfected with either miR-29b or NC and co-cultured with U266 MM cells. 24 hours after transfection, cell were harvested and separated with immuno-magnetic microbeads.
Growth protocol DCs were generated from healthy donors' CD14+ monocytes isolated by immunomagnetic separation. These cells were then cultured for 6 days in RPMI-1640 medium containing 10% FBS, 1% penicillin/streptomycin, 100 ng/ml recombinant human GM-CSF and 50 ng/ml IL-4 . Half-medium was replaced every 48 hours.

Extraction protocol total RNA (tRNA) from DCs was extracted through column purification with RNeasy kit (Qiagen, Hilden, Germany)
Label biotin
Label protocol cDNA was processed for fragmentation and labeling using the Affymetrix® GeneChip® WT Terminal Labeling Kit (Affymetrix Inc.) according to the Affymetrix’s procedures
 
Hybridization protocol cDNA was hybridized on GeneChip® Human Transcriptome Array 2.0 at 45C for 16-18 hours. The arrays were washed and stained on Affymetrix Fluidics Stations 450.
Scan protocol The arrays were scanned with an Affymetrix GeneChip Scanner 3000

Data processing Data were processed using Affymetrix Expression Console for quality control after summarization and Robust Multi-array Average (RMA) normalization
