ES cells are cultured in gelatin wells with feeder free ES cell medium: 1 x GMEM medium supplemented with 2 mM glutamine, 1 mM sodium pyruvate,1 x nonessential amino acids,10% (v/v) fetal bovine serum,a 1:1000 dilution of beta-mercaptoethanol stock solution, and 500-1000 units per ml of leukocyte inhibitory factor.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 8 microg total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Following fragmentation, 15 microg of cRNA were hybridized for 16 hr at 45C on GeneChip mouse genome 430 2.0. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

GeneChips were scanned using the Affymetrix GeneChip scanner.

Data processing |
The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 100.
