Title NK cells, 40 ng/mlTGF-b1_donor3
Sample type RNA

Source name Blood, NK cells
Organism Homo sapiens
Characteristics tissue: blood
cell type: NK cells
Extracted molecule total RNA
Extraction protocol Total RNA containing the small RNA fraction was extracted using the miRCURY RNA Isolation Kit - Cell and Plant (Exiqon)
Label FAM
Label protocol Total RNA was reverse transcribed using the TaqMan MicroRNA Reverse Transcription kit with the Megaplex RT Primers Human Pool A+B set v.3.0 (catalog number 4444913, Applied Biosystems), a preamplification step was performed using the TaqMan PreAmp Master Mix with the Megaplex PreAmp Primers Human Pool A+B set v.3.0, (Applied Biosystems) preamplified cDNAs were loaded in TaqMan Array Human MicroRNA Cards A and B set v3.0 (Applied Biosystems).

Hybridization protocol n/a
Scan protocol n/a
Description Test
Data processing Data were analyzed using the miScript miRNA PCR Array Data Analysis Tool (http://www.sabiosciences.com/mirnaArrayDataAnalysis.php). MiRNAs expression was normalized to the U6 snRNA expression, for which four replicate wells are present in each array card. The ΔΔCt method was used. The web-based software automatically performed all calculations from the uploaded raw threshold cycle data.
normalized data file reports normalized signal (against reference U6 snRNA). Fold Change file reports test/control (i.e., treated NK/untreated NK) ratios.
