Title |
WT_Colon_MF_Rep2 |
Sample type |
RNA |
Source name |
Colon, isolated MF, Replicate 2
Organism |
Mus musculus |
Characteristics |
tissue specific cells: Colon macrophages gender: male age: 6 weeks
Treatment protocol |
Cells were isolated ex vivo and cells were directly isolated within a total range of time of 5h.
Growth protocol |
Different macrophage populations were freshly isolated from WT mice (C57BL6/J) grown in SPF conditions.
Extracted molecule |
total RNA |
Extraction protocol |
The miRNeasy total RNA extraction kit (Qiagen) was used for RNA isolation.
Label |
Cy3
Label protocol |
Cyanine-3 (Cy3) labeled miRNA was prepared from 100ng total RNA using Agilent miRNA Complete Labeling and Hyb Kit according to the manufacturer's instructions
Hybridization protocol |
Labeled RNA was hybridized with a 8X15 mouse miRNA array rel v19 (design number 046065) at 55°C and 20 rpm for 20 hours, and then washed according to the manufacturers instructions.
Scan protocol |
Slides were scanned immediately after washing on Agilent Technologies Scanner G2505B US22502692 using 5uM resolution.
Description |
miRNA expression in colon macrophages
Data processing |
The scanned images were analyzed with Feature Extraction Software 9.1 (Agilent) using default parameters (protocol miRNA_107_Sep09 and Grid: 029152_D_F_20100702) to obtain background subtracted and spatially detrended Processed Signal intensities.
