tissue: For the preparation of cancer-associated fibroblasts (CAF) and benign/normal associated fibroblasts (NAF), de-identified prostate tissue samples were collected at Vanderbilt University under local IRB approval through the NCI-funded Cooperative Human Tissue Network. CAF and NAF were isolated from cancer and normal tissue respectively, as previously described (PMID:11719442). CAF activity was validated using a tissue recombination bioassay in which CAF or NAF were recombined with initiated reporter epithelium (BPH-1) and grafted to the renal capsule of severe combined immunodeficient (SCID) mice. Cells were classified as CAF if they demonstrated the ability to induce tumorigenesis (defined grossly and histologically) in the epithelial cells under these conditions. Cells from benign sources were also checked to confirm that they did not induce tumor growth. Genomic DNA was extracted from cells using the QIAamp DNA mini kit (Qiagen, Inc.) according to the manufacturer’s instructions.
