Growth protocol All the EP300 knocked-down and their corresponding paclitaxel resistant derivatives MCF7 cells were grown in Dulbecco's Modified Eagle's Medium (DMEM) containing fetal calf serum (FCS) and glutamine.
Extracted molecule total RNA
Extraction protocol RNA extractions were performed using RNeasy Mini Kit from QIAGEN and according to the manufacturer's instruction. A post-extraction QC checking of the RNA sample integrity was performed by measuring RNA Integrity Number (RIN) using a bioanalyser (Agilent 2100, DE24802458, ver. 2.6).
Label biotin
Label protocol Biotinylated single-stranded cDNA (ss-cDNA) samples were prepared according to the standard Affymetrix protocol from 5.5 µg ss-cDNA (User Guide for GeneChip® WT PLUS Reagent Kit, Manual Target Preparation for GeneChip® Whole Transcript (WT) Expression Arrays (P/N 703147 Rev. 3, Affymetrix).
 
Hybridization protocol Following fragmentation and labeling, 3.5 µg of ss-cDNA were hybridized for 16 hr at 45°C on GeneChip® Cartridge Array. GeneChips were washed and stained in the GeneChip® Fluidics Station 450 (Affymetrix, P/N 00-0079).
Scan protocol GeneChips were scanned using the GeneChip® Scanner 3000 7G.

Data processing The data were analyzed with Partek Genomics Suite version 6.6 using Affymetrix default analysis settings and global scaling as normalization method.
