Treatment protocol |
Immediately following death, tongue were excised, longitudinally bisected, each half of the tongue was frozen on dry ice, embedded in OCT and stored at -80°C. Epithelial cells were isolated using an infrared laser microdissection system (Arcturus PixCell II®, Life Technologies, NY, USA). Microdissected cells on the cap were solubilized using a RLT lysis buffer and were subjected to total RNA extraction.

Total RNA was performed using the Rneasy Microkit® (Qiagen) according to the manufacturer's instructions.

Following amplification of 2ng of RNA (Ovation Pico WTA System v2, NuGEN Technologies), each amplified sense cDNA product was fragmented and biotinylated, using an Encore Biotin Module according to manufacturer’s instructions (NuGEN Technologies).
