Growth protocol |
Mononuclear cells from PB and LN single-cell suspension were isolated by density gradient centrifugation using the lymphocyte separation medium (ICN Biomedicals). Fresh cells were subject to CD19 selection by magnetic beads labeling using the MACS columns (Miltenyi Biotec). CD19 samples with purity of >96% were stored at -80°C for subsequent use.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 ug total RNA (Expression Analysis Technical Manual, Affymetrix).

Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Drosophila Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.

GeneChips were scanned using the Hewlett-Packard GeneArray Scanner G2500A.
