Total RNA was extracted using the protocol of Chang et al. (1993) Plant Mol. Biol. Rep. 11: 113-116. Small RNAs were then purified by PAGE, reverse-transcribed, and sequencing using pyrosequencing (454 Life Sciences) as previously described by Axtell et al. (2006) Cell 127:565-577.
