The culture conditions described previously by Vasconcelos et al. (Vasconcelos I, et al. J Bacteriol. 1994; 176, 1443-1450.) and Girbal et al. (Girbal L, Soucaille P. J Bacteriol. 1995; 176,6433-6438.) were used for the phosphate-limited continuous culture of C. acetobutylicum, fed with a constant total carbon amount of 995 mM. The cultures were maintained under acidogenesis (pH 6.3, 995 mM of carbon from glucose), solventogenesis (pH 4.4, 995 mM of carbon from glucose) and alcohologenesis (pH 6.3, 498 mM of carbon from glucose and 498 mM of carbon from glycerol).

Extraction protocol |
RNA was extracted using an RNeasy Midi kit (Qiagen, Courtaboeuf, France) following the manufacturer's instructions with supplementation of DNase treatment using RNase-Free DNase Set (Qiagen). RNA quantity and quality evaluations were performed using an Agilent 2100 Bioanalyzer (Agilent Technologies, Massy, France) and a NanoDrop ND-1000 spectrophotometer (Labtech France, Paris, France) at 260 nm and 280 nm.
