| Treatment protocol |
Pairs of fetal ovaries/post-natal testes were dissected from embryos/mouse that were previously weighed. Gonads were dissolved in 50µl RLT lysis buffer (Qiagen) and stored at -20°C before pools constitution. For males, testes were weighed and contralateral testes were fixed in Formol for histological analysis.

Mice were breeded in conventional animal facility in accordance with animal health care, ethics and current regulations.

Gonads in RLT buffer were pooled and lysed with homogenizer before RNA extraction using RNeasy Mini kit (Qiagen) according to the manufacturer's instructions.

Biotinylated cDNA were prepared according to the standard Affymetrix protocol from total RNA (following the Affymetrix GeneChip Whole Transcript (WT) Sense Target Labelling Assay kit).

Following fragmentation, cDNA were hybridized for 16 hr at 45C on Affymetrix Mouse Gene 2.0 ST Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

GeneChips were scanned using the GCS 3000 scanner (Affymetrix).
