Transgenic Ler seedling carrying GRREV transgene. Transgene is a fusion of glucocorticoid receptor to Arabidopsis REVOLUTA protein. Expression is driven by 35S promoter. Seedlings were 10 days old.

Treatment protocol |
Seedlings were treated with dexamethasone in ethanol to a final concentration of 0.1% ethanol and 50 micrograms per ml dexamethasone for one hour before tissue was harvested and quick frozen.

Growth protocol |
Seedlings were grown in liquid culture, in 50 ml MS salts plus glucose in 250 ml flasks, 100 seeds per flask. Flasks were shaken gently on a rotator under fluorescent lights (24 hour, 22 degrees). Seeds were stratified at 4 degrees in water for 2 days prior to adding to culture medium.
