Characteristics Transgenic Ler seedling carrying GRREV transgene. Transgene is a fusion of glucocorticoid receptor to Arabidopsis REVOLUTA protein. Expression is driven by 35S promoter. Seedlings were 10 days old.
Treatment protocol Seedlings were treated with dexamethasone in ethanol to a final concentration of 0.1% ethanol and 50 micrograms per ml dexamethasone for one hour before tissue was harvested and quick frozen.
Growth protocol Seedlings were grown in liquid culture, in 50 ml MS salts plus glucose in 250 ml flasks, 100 seeds per flask. Flasks were shaken gently on a rotator under fluorescent lights (24 hour, 22 degrees). Seeds were stratified at 4 degrees in water for 2 days prior to adding to culture medium.
Extracted molecule total RNA
Extraction protocol RNA was isolated from 100 mg of tissue using a QIAGEN RNeasy plant miniprep kit.
Label biotin
Label protocol Twenty micrograms biotinylated cRNA was prepared from the purified total RNA using standard Affymetrix protocols.
