Sample GSM154092 Query DataSets for GSM154092
Status Public on May 16, 2007
Title MycFlag-G1-arrest-t150-rep2
Sample type other

Channel 1
Source name Myc IP 150 minutes rep2
Organism Saccharomyces cerevisiae
Characteristics Strain: MDY510
Treatment protocol Cells arrested with 4μg/mL alpha factor for 3 hours, collected by filtration and resuspended in YP+2% galactose.
Growth protocol MDY510 cells were grown in YP + 2% raffinose
Extracted molecule other
Extraction protocol IPed using anti-Myc Ab (Sigma; monoclonal). Amplified with a starting amount of up to 75 ng for ChIP samples, using the DNA linear amplification
Label Cy5
Label protocol 3 μg of aRNA produced from the linear amplification were used to label probe

Channel 2
Source name Flag IP 150 minutes rep2
Organism Saccharomyces cerevisiae
Characteristics Strain: MDY510
Treatment protocol Cells arrested with 4μg/mL alpha factor for 3 hours, collected by filtration and resuspended in YP+2% galactose.
Growth protocol MDY510 cells were grown in YP + 2% raffinose
Extracted molecule other
Extraction protocol IPed using anti-Flag Ab (Sigma; monoclonal). Amplified with a starting amount of up to 75 ng for ChIP samples, using the DNA linear amplification
Label Cy3
Label protocol 3 μg of aRNA produced from the linear amplification were used to label probe

Hybridization protocol Labeled probes were hybridized onto a yeast tiled oligonucleotide microarray [Yuan et al., Science 309, 626 (2005)] at 65°C for 16 hours and washed as described on www.microarrays.org.
Scan protocol The arrays were scanned at 5 micron resolution with an Axon Laboratories GenePix 4000B scanner running GenePix 5.1. Image analysis and data normalization were performed as previously described [Liu et al., PLoS Biol 3, e328 (2005)].
Description High-resolution Myc/Flag rations (using IP for Gal-driven Flag-H3 and constitutive Myc-H3). 150min after Gal induction.
