Human Hematopoietic Progenitors (hHP; CD34+) were purified using immunomagnetic beads (Miltenyi Biotech, Germany) from human umbilical cord blood (CB). They were cultured in Iscove’s modified Dulbecco’s media (IMDM) containing 20% BIT 9500 Serum substitute (STEMCELL Technologies), 1% penicillin-streptomycin (Invitrogen), 0.2% β-mercaptoethanol, 10 ng/ml SCF, megakaryocyte growth and development factor (TPO), FLT3 ligand (FLT3L), IL-3, and IL-6.
