Treatment protocol |
Licochalcone A stock solution, prepared in dimethyl sulfoxide (DMSO), was added to the cultures, giving a final concentration of 1/2× MIC (2 μg/ml), and the final concentration of DMSO was 0.1% (vol/vol) as control group. Biofilms washed once with PBS to remove any unbound bacteria, Licochalcone A stock solution, prepared in dimethyl sulfoxide (DMSO), was added to the cultures, giving a final concentration of 4× MIBC (64 μg/ml), and the final concentration of DMSO was 0.1% (vol/vol) as control group. Then, the bacterial planktonic and biofilm cells were further incubated for 60 min at 37°C

Growth protocol |
S. aureus strain ATCC29213 was grown overnight at 200 rpm in a rotary shaker at 37°C in 10 ml of TSB, after 12 hours, the culture were inoculated to an initial OD600 of 0.05 in 100 ml of TSB in 250-ml Erlenmeyer flasks, the planktonic cells were grown at 37°C at 200 rpm to an OD600 of 0.3. Within biofilm, the overnight culture were inoculated to an initial OD600 and diluted in TSBg to obtain the final concentration of 1× 105cfu/ml, 30 ml bacterial suspension diluted in 2% glucose-supplemented TSB containing 1 × 105 cfu/ml into the culture dish, then incubated in air for 48 h at 37°C
