Treatment protocol Cells were transfected with control or SRC-1 targeting siRNA using DharmaFECT 1 according to the manufacturer's protocol. Transfected cells were changed to medium supplemented with 10% dialyzed FCS and incubated for 36 hours before the indicated treatment with DMEM with or without glucose supplemented with 10% dialyzed FCS.
Growth protocol A549 cell lines were cultured in DMEM supplemented with 10% fetal calf serum (FCS), penicillin, and streptomycin (100 U/mL). Twenty-four hours prior to transfection, A549 cells were plated in 6-well plates.
Extracted molecule total RNA
Extraction protocol RNAs was isolated using the RNeasy mini Kit (QIAGEN).
Label biotin
Label protocol Biotinylated cRNA was produced using the standard Affymetrix protocol.
 
Hybridization protocol 15 ug of cRNA were hybridized for 16 hr at 45 degrees C on GeneChip Genome Array. GeneChips were washed, then stained in the Affymetrix Fluidics Station 450.
Scan protocol GeneChips were scanned with the GeneChip Scanner 3000.

Data processing The data were analyzed using RMAExpress Version: 1.0.4 with default analysis settings.
