Treatment protocol |
Cells were incubated in phenol red free medium for 4hr (to ensure complete removal of phenol red) and treatment was started, after a DPBS wash, by adding fresh phenol red free medium along with appropriate drug or vehicle.

Growth protocol |
MCF7 cells were maintained in phenol red containing DMEM-F12 supplemented with 10%FBS. For experiments cells were seeded in 6 well plates at a density of 2*10^5 cells/well in phenol red containing medium. After 48hr spent medium was removed, cells were washed with DPBS and phenol red free DMEM-F12, supplemented with 10% charcoal stripped dextran treated FCS, was added.

After the After 24hr of treatment the spent medium was removed and cells were lysed in RLT lysis buffer and total RNA was isolated by Qiagen’s RNeasy minikit.

Data processing |
Data processing was done in R software environment using LIMMA package. Raw median signal intensities were background subtracted by normexp method and between arrays normalization was done by quantile method. Probe ID replicates were averaged. Lmfit and eBayes (Empirical Bayes) methods were used to identify differentially regulated genes.
