Treatment protocol SU5402 incubation was performed as previously described (Marques et al., 2008). A 1 mM stock of SU5402 (Calbiochem) in DMSO was diluted to a working concentration of 10 μM in E3 medium. Up to 20 embryos were treated in glass vials in a final volume of 3 mL. Control embryos were treated with 1% DMSO in E3 medium.
Growth protocol Embryos were collected, grown in blue water (60 mg/mL Instant Ocean and 0.025% methylene blue) at 28.5 degrees Celcius and dechorionated prior to heart dissection and RNA extraction.
Extracted molecule total RNA
Extraction protocol Embryonic zebrafish hearts and surrounding tissues were collected at 48 hpf for microarray analysis using a previously described syringe-based extraction method (Burns and MacRae, 2006). Tissue was preserved in RNAlater solution (Ambion) and total RNA was isolated using a RNAqueous-Micro kit (Ambion).
