Treatment protocol |
PBMC was isolated from peripheral whole blood using Ficoll-Plaque Plus (Amersham Biosciences, USA) according to the manufacturer’s protocol. In brief, one ratio of diluted blood (1:1 ratio in PBS) sample was layered on 2 ratio of Ficoll-Plaque Plus (Amersham Biosciences, USA), pelleted (400g, 30 minutes at 18-20°C) and the buffy coat was collected, washed twice with PBS. PBMC with viability higher than 90% was subjected to RNA extraction.

Growth protocol |
Peripheral whole blood was collected in heparinized vacutainer tubes (Becton Dickinson) and subjected to PBMC isolation within 4 hours.

Following fragmentation, 12.5 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

Data processing |
The data value(log2) were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method.
