Saccharomyces cerevisiae |
Characteristics |
strain_id: JG1830 genotype/variation: fob1 mutant replicate: 2 experiment_date: Jun2011
Treatment protocol |
Cells growing in exponential phase in YPD+CSM were pelleted by centrifugation.
Growth protocol |
Cultures were innoculated from fresh overnights and grown at 30C in YPD+CSM to an OD as indicated.
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was extracted using the hot acid phenol method.
Label |
biotin
Label protocol |
Biotinylated cRNA was prepared from 300 ng Total RNA using MessageAmp III aRNA Kit from Ambion.
Hybridization protocol |
Following fragmentation, 5 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Yeast Genome 2.0 Arrays. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.
Scan protocol |
GeneChips were scanned using the Hewlett-Packard GeneArray Scanner G2500A.
Description |
fob1_2.2
Data processing |
The Affymetrix Yeast Genome 2.0 array contains probes designed for both S. pombe and S. cerevisiae. Data from the arrays was filtered to exclude probes annotated to S. pombe, and retain those annotated to S. cerevisiae genes. Array intensities were processed using the RMA algorithm in the affy library (version affy_1.36.1) from bioconductor in R version 2.15.2.
