Treatment protocol |
For the first developmental stage, embryonic day (E) 7.5, whole embryos were collected. For later stages cardiac structures were micro-dissected including E8.5 heart tubes and left and right ventricles corresponding to E9.5, E12.5, E14.5, E18.5, newborn and adult mice. All samples were harvested in biological triplicates, with approximately 30-50 embryos/hearts included per replicate corresponding to E7.5, E8.5 and E9.5; 10-20 hearts for each E12.5 or E14.5 sample; 3-5 hearts pooled for each E18.5 replicate and one used for newborn and adult samples. Embryonic stem cells (ESC) from the R1 line were used as pluripotent gold standard. To avoid interference due to high protein content newborn and adult hearts were fast-frozen and pulverized prior to RNA extraction.

Growth protocol |
Wild type CD1 strain mouse embryos were harvested at defined stages from timed mating pairs. Morphology and somite count were used as inclusion criteria for each developmental time point.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 100 ng total RNA.

Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on Affymetrix Mouse Genome 430 2.0 Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

Scan protocol |
GeneChips were scanned using the GeneChip Scanner 3000 7G.
