Cells were transfected with siRNAs and stimulated with 3 ng/ml of TGF-beta for the indicated times.

Growth protocol |
H441 cells were maintained in RPMI 1640 (#11875; Life Technologies) supplemented with 10% FBS, 100 U/ml penicillin G, and 100 ug/ml streptomycin. Cells were grown in a humidified atmosphere with 5% CO2 at 37°C.

Double-stranded cDNA was synthesized with the SuperScript Choice system (GIBCO) and a T7-(dT) 24 Primer (GE Amersham) from the total RNA. In vitro transcription was performed to produce biotin-labeled cRNA by using a BioArray HighYield RNA Transcript Labeling Kit (Affymetrix) according to the manufacturer’s instructions (Ishii et al, Genomics 68:136-143, 2000).

Microarray Suite software 5.0 (Affymetrix) was used to calculate the average difference (AD) for each gene probe set, which was shown as an intensity value of the gene expression (Ishii et al, Genomics 68:136-143, 2000).
