Treatment protocol |
Within 6 hours of collection, blood was processed by density gradient centrifugation using Ficoll-Paque PLUS (GE Healthcare, Sweden). Residual red blood cells were lysed and washed with phosphate-buffered saline (PBS). The isolated peripheral blood mononuclear cells (PBMCs) were stored at -80 oC.

Growth protocol |
Patients with primary pancreatic, gastric or HCC were recruited at diagnosis. Informed consent was obtained from all subjects before blood samples were collected. A total of 10 ml of blood was collected into BD Vacutainer® Plus K2EDTA tubes (Becton-Dickinson, USA).

Extraction protocol |
Total RNA was extracted from PBMC and WBC using TRIzol reagent (Invitrogen, USA) according to the manufacturer's protocol.

Five micrograms of total RNA were reverse-transcribed and labeled with biotin.

The final cRNA obtained was hybridized to the GeneChip Human Genome U133 Plus 2.0 Array (Affymetrix, USA).

GeneChips were scanned using the Affymetrix GeneChip Scanner 3000.

Data processing |
The data was analysed using Partek Genomics Suite version 6.6. CEL files were imported into Partek with GCRMA background correction, quantile normalisation and log2 transformation.
