First and second strand complimentary DNA was synthesized using standard techniques. Biotin-labeled antisense complimentary RNA was produced by an in vitro transcription reaction.
Hybridization protocol
Mouse Genome 430 2.0 microarrays (Affymetrix, Foster City, CA) were hybridized with 20 µg cRNA. Target hybridization, washing, staining, and scanning probe arrays were done following an Affymetrix GeneChip Expression Analysis Manual.
