Growth protocol |
Mice were bred and kept at the GSF animal facilities in isolated ventilated cages type II with wood shavings (Altromin, Lage, Germany) as bedding at a temperature of 20 to 24o C, humidity of 50 to 60%, 20 air exchanges per hour and a 12/12-hour light/dark cycle. They were tested for microorganisms in 12-week intervals according to the FELASA Guidelines to the level required for rederived mice

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 3 microg total RNA (Expression Analysis Technical Manual, 2004, Affymetrix).

Following fragmentation, 10 microg of cRNA were hybridized for 16 hr at 45C on GeneChip Mouse Genome 430 2.0 Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

GeneChips were scanned using the Affymetrix GeneChip Scaner 3000.

Data processing |
Genewise tests for significantly regulated transcripts between creatine treated and untreated mice were performed in the statistical programming language R with BioConductor using the dChip and the Robust Multichip Average (RMA) probeset summaries. Data are log-transformed and normalized with a lowess-smoother operating on MA-scale against the mean of the creatine treated group. Transcripts are called significantly regulated if the corresponding p-value of the Welch's t-test is below the 5% FDR significance level.
