Treatment protocol CD4+ T cells of each approach were pooled, counted and resuspended in TRIzol reagent.
Growth protocol CD4+ T cells were prepared from 2 male and 2 female donors, enriched by negative selection (BD IMag) and cultured at 37°C, 5% CO2 for 30 h.

Extraction protocol RNA was isolated by chloroform/isopropanol extraction. For RNA concentration and purification the RNeasy MinElute Cleanup Kit (Qiagen) was used as recommended by the manufacturer.

Label protocol Reverse transcription of RNA to synthesize cDNA was done by using the WT Expression Kit (Ambion). cDNA fragmentation and labeling was performed by applying the GeneChip WT Terminal Labeling Kit (Affymetrix).
 
Hybridization protocol Probes were prepared using the GeneChip Hybridization, Wash, and Stain Kit (Affymetrix) and analyzed by microarray technology (Affymetrix GeneChip Human Gene 1.0 ST Array).
Scan protocol The gene-expression scan was performed with the GeneChip Command Console 3.0 Software (Affymetrix).

Gene expression data from CD4+ T cells
Data processing The robust multi-array average (RMA) algorithm was applied for background correction, using the Expression Console 1.1 Software (Affymetrix). The samples were quantile normalized, followed by mean signal summarization.
