Treatment protocol Ureters at E14.5 and E16.5 were microdissected and placed in RNAlater solution (Ambion, Austin, TX) until sufficients samples had been collected to perform the experiments in triplicate for each condition.
Extracted molecule total RNA
Extraction protocol Total RNA was isolated from ureters using TRIsure Reagent (Bioline) according to manufacturer's instructions. The integrity of RNA was assessed using an Agilent 2100 Bioanalyser (Agilent Technology) and RNA concentration determined using a NanoDrop ND-1000 spectrophotometer (NanoDrop, Rockland, DE).
Label biotin
Label protocol cRNAs for hybridisation to Affymetrix arrays were prepared from 100 ng total RNA using the wild-type expression kit Affymetrix (Affymetrix, Santa Clara, CA 95051).
 
Hybridization protocol Labelled-cRNA was fragmented and hybridised to mouse gene 1.0 ST arrays (Affymetrix) following manufacturer’s protocols. Three independent RNA preparations from the four different conditions [wild type and mutant, E14.5 and E16.5] were processed and hybridised on Mouse Gene Arrays.

Description Gene expression data from E14.5 ureter
Data processing Arrays have been quantile-normalize with respect to the probe GC content using the RMA algorithm (GC contend adjustment, RMA background correction and mean probe set summarization). Not or low expressed transcripts have been removed by a maximum expression cutoff < 100.
