Treatment protocol |
150g male Wistar rats (Harlan) were injected with 65 mg/kg streptozotocin (STZ) (Diabetic) or citrate buffer (Control). Ventricles were collected and frozen after 3/28/42 days, corresponding to diabetes response with no symptoms, diastolic symptoms, and full diastolic and systolic symptoms of diabetic cardiomyopathy.

Rats were kept separate in steel cages with full access to standard feed and water. Ventricles removed at 03/28/42 day time points, frozen and total RNA extracted by Trizol method

Trizol extraction of total RNA was performed according to the manufacturer's instructions.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 microg total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Hybridization protocol |
Following fragmentation, 10 microg of cRNA were hybridized on Affymetrix GeneChip Rat genome RG-U34A. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.

Scan protocol |
GeneChips were scanned using the Hewlett-Packard GeneArray Scanner 3000.

Data processing |
The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 100.
