Title |
keratinocytes_anti-miR-Ctrl_rep1 |
Sample type |
RNA |
Source name |
keratinocytes transfected with anti-miR-Ctrl
Organism |
Homo sapiens |
Characteristics |
tissue: skin cell type: keratinocytes hairpin rna: anti-miR-Ctrl
Treatment protocol |
The third passage keratinocytes at 30-50% confluence were transfected with 10 nM miR-31 hairpin inhibitor or microRNA hairpin inhibitor negative control #1 (ThermoFisher Scientific) using Lipofectamine 2000 (Invitrogen).
Growth protocol |
Human adult epidermal keratinocytes were purchased from Cascade Biologics and cultured in EpiLife® serum-free keratinocyte growth medium including Human Keratinocyte Growth Supplement at a final Ca2+ concentration of 0.06mM (Cascade Biologics) at 37°C in 5% CO2.
Extracted molecule |
total RNA |
Extraction protocol |
48 hours after transfection, the total RNA was extracted with the miRNeasy Mini kit (Qiagen) according to the manufacturer's instructions.
Label |
biotin
Label protocol |
Total RNA was labeled using the Whole Transcript (WT) Sense Target Labeling Assay according to the manufacturer's (Affymetrix) instructions.
Hybridization protocol |
The samples were hybridized to a Human Gene 1.1 ST Array Plate according to the standard Affymetrix GeneTitan protocol.
Scan protocol |
The array plate was scanned using the Affymetrix GeneTitan Scanner.
Description |
Anti-miR-Ctrla Gene expression data from keratinocytes transfected with control RNA with scrambled sequence.
Data processing |
Generated CEL files were analyzed using Affymetrix Expression Console v. 1.1 (Summarization: Plier, Background Correction: PM-GCBG, Normalization: Global Median).
