Summary |
Doxorubicin (DOX) and other anthracyclines are effective chemotherapeutic agents, however, their use is influenced by the risk of cardiotoxicity. We still have an incomplete understanding of the cardiomyocyte protective pathways activated after anthracycline-induced cardiotoxicity (AIC).Danshen injection (DSI), astaxanthin (AXT) and diosmetin (DMT) are effective in the treatment of cardiovascular diseases, but the mechanism of protection against adriamycin-induced cardiotoxicity is unclear. Here, we performed RNA-seq screening in H9c2 cardiomyocytes to determine the potential protective mechanisms of Danshen injection, astaxanthin and diosmetin against AIC.

Overall design |
To investigate the therapeutic effects of danshen injection, astaxanthin and diosmetin, respectively, in anthracycline-induced cardiotoxicity, we divided cultured H9c2 cells into five groups:a control group treated with PBS, a model group exposed to 2μM DOX, a treatment group subjected to 2μM DOX in combination with 50μL DSI diluted 1/20 in culture volume, a treatment group subjected to 2μM DOX in combination with 5μM AXT and a treatment group subjected to 2μM DOX in combination with 10μM DMT. Each group consisted of three samples, with DOX added after 4 hours of DSI, AXT an DMT treatment. The cells were harvested 24 hours after DOX treatment, then we performed gene expression profiling analysis using data obtained from RNA-seq.
