Summary |
m6A mRNA methylation in brown adipose tissue regulates systemic insulin sensitivity. We performed N6-methyladenosine (m6A) profiling and RNA seq of brown adipose tissue (BAT) from control and Mettl14-KO mice. Meanwhile, we performed RNA-seq for control and Mettl14-KO human BAT cell line as well.

Overall design |
The experiment was performed in homozygous, male control, and M14 KO mice. All mice were fed with chow-diet for twenty-four weeks from four week of age. At the end of the experiment, mice were euthanized after overnight fasting, followed by serum and tissue sample collection. hBAT cells were differentiated for 21 days, washed with PBS and cultured in 1% BSA containing high-glucose DMEM medium for 24 hours, and then collected cell pellet for RNA extraction.
