Title mRNA-seq in VDR knockout and control THP-1 cells

Summary The vitamin D receptor (VDR) has been knocked out in monocytic THP-1 cells after stimulation of ko and control cells with the the natural VDR ligand 1,25-dihydroxyvitamin D3 (1,25(OH)2D3), quadruplicate mRNA-seq has been performed
 
Overall design The knockout has been performed by lentiviral CRISPR/Cas9 gene editing using a guideRNA (gRNA) directed against the VDR gene. The control cells have been created using a non-targeting gRNA (NTC). Prior to MRNA-seq, the cells were treated for 24 h with 100 nM 1,25(OH)2D3 or the solvent control (EtOH).
