Overall design For the ATAC-seq analyses of Y and O HSCs pre- and post-transplantation, we utilized a paired approach. For each replicate, pools of 3 to 4 Y B6 mice and 1 O B6 mouse were used to isolated 10,000 pre-transplantation HSCs for ATAC-seq sample preparation and to transplant 1,000 HSCs into 5 lethally-irradiated Y BJ recipient mice alongside 300,000 Sca-1 depleted BJ helper BM cells. Post-transplantation samples were collected 3 months after transplantation from the pooled BM of the 5 recipient mice to obtain 10,000 post-transplantation HSCs for ATAC-seq sample preparation.
