Summary |
We thus isolated cortex from mice, with or without sevoflurane exposure, and performed single cell sequencing. Single cell sequencing is a innovative experimental procedures which allows the identification of various subsets of cells in the neverous system after sevoflurane exposure. Isolated cells were concentrated to 1000 cells/uL and approximately 17,000 cells were loaded into each channel to generate single-cell Gel Bead-In-Emulsions (GEMs), which led to expected mRNA barcoding of 10,000 single-cells for each sample. Seurat package was used for cell normalization and regression. Sevoflurane and control groups were aligned and combined using Seurat. Using t-distributed stochastic neighbor embedding (tSNE), 13 cluters of cells were visualized. Cells were assigned to various clusters based on strong expression of specific genes.
