Summary |
We used human foreskin fibroblast cells with CRISPR-Cas9 mediated knockout of RB1 and p130 (RBL2) (Control, sgRB1, sgP130, sgRB1+sg130) and measured gene expression changes after doxorubicin treatmnet (24 hr, 350 nM).

Overall design |
Control, sgP130, sgRB1, and sgRB1+sgP130 HFFs were contact arrested for 48 hours. Cells were released from contact inhibition by splitting into complete media containing 0 nM or 350 nM doxorubicin. Cells were collected after 24 hours and RNA was harvested for sequencing. Experiment was completed with two biological replicates for each cell line in each condition resulting in 16 samples run in a single lane for RNAseq.
