Transcription factor Pax5 overexpression in Hodgkin lymphoma cell line L428

Analysis of classical Hodgkin lymphoma (cHL) cell line L428 stably-transfected with a mifepristone-inducible PAX5-expression system. The L428-PAX5 cells were epigenetically modified by 5-aza-2'-deoxycytidine/TSA treatment. Results provide insight into the role of PAX5 in cHL.

Expression data from Hodgkin lymphoma cell lines L-428 and L-1236: L-428 and L-1236 cells were transduced with tGFP-EBF1 or as control tGFP-only.

(Submitter supplied) The transcription factor network in Hodgkin lymphoma (HL) represents a unique composition of proteins found in no other hematopoietic cell. An aberrant downregulation of the B cell transcription factor EBF1 is observed in the B cell-derived Hodgkin and Reed/Sternberg (HRS) tumor cells. Herein, we elucidated the consequences of the down regulation of this factor in HL. To obtain a more comprehensive overview of EBF1 regulated genes in cHL cell lines, we performed a gene chip analysis comparing gene expression in tGFP-EBF1-transduced L-1236 and L-428 cells compared to samples transduced with vectors encoding only tGFP.

Epigenetic upregulation of B-cell inappropriate genes induces extinction of B-cell program in classical Hodgkin lymphoma

Chip-chip from Z138 MCL cell line with SOX11 antibody

(Submitter supplied) To determine the role of SOX11 in aggressive lymphoid neoplasm, we first investigated direct target genes of this transcription factor using chromatin immunoprecipitation and DNA microarrays (ChIP-chip) in MCL cell lines. Analysis of the exact location of SOX11 binding elements showed that SOX11 interacts with DNA predominantly in the enhancer regions (in 40% of the enriched regions) and intron segments (in 33% of the enriched regions), suggesting that in these genes SOX11 may interact with the general transcriptional machinery and transcriptional co-regulators.

Ebf1 or Pax5 Haploinsufficiency Synergizes with STAT5 Activation to Initiate Acute Lymphoblastic Leukemia

(Submitter supplied) STAT5 is critical for differentiation, proliferation and survival of progenitor B cells suggesting a possible role in Acute Lymphoblastic Leukemia (ALL). Germline Aberrations of PAX5 Cause Susceptibility to pre-B cell Acute Lymphoblastic Leukemia

(Submitter supplied) A novel heterozygous germline variant, c.547G>A (p.Gly183Ser), in the paired box protein encoding gene, PAX5, was found to segregate with disease in two unrelated kindreds with autosomal dominant pre-B cell acute lymphoblastic leukemia (ALL).
