Suspension cell cultures response to overexpression of master regulators VND6 and SND1

Analysis of suspension cell lines in which VASCULAR-RELATED NAC-DOMAIN6 (VND6) or SECONDARY WALL-ASSOCIATES NAC DOMAIN PROTEIN1 (SND1) was overexpressed after the addition of estrogen. Results provide insight in to the roles of master regulators VND6 and SND1 in xylem development.

Expression data from Arabidopsis suspension cells overexpressing VND6 and SND1

(Submitter supplied) Xylem consists of three types of cells: vessel cells, also referred to as tracheary elements (TEs), parenchyma cells, and fiber cells. TE differentiation includes two essential processes, programmed cell death (PCD) and secondary cell wall formation. These two processes are tightly coupled. However, little is known about the molecular mechanism of their gene regulation. Here, we show that VASCULAR-RELATED NAC-DOMAIN 6 (VND6), a master regulator of TEs, regulates these processes in a coordinated manner. more...

Gene expression over the course of xylem tracheary element formation in cell suspension cultures of Arabidopsis thaliana

(Submitter supplied) Transcriptomic analysis of gene expression during the differentiation of cell suspension cultures into tracheary elements using the biological system published by Pesquet et al., Current Biology (2010): tracheary element differentiation was triggered by externally supplying hormone-free habituated cell suspension cultures of Arabidopsis thaliana Col-0 with auxin, cytokinin and epibrassinolides; RNA samples extracted from 3 independent time-courses every 12h from 0h to 4 days were analyzed using ATH1 Arabidopsis Affymetrix micro-array

Finding direct target genes of VND7

(Submitter supplied) The Arabidopsis thaliana NAC domain transcription factor, VASCULAR-RELATED NAC-DOMAIN7 (VND7), acts as a key regulator of xylem vessel differentiation. In order to identify direct target genes of VND7, we performed global transcriptome analysis using Arabidopsis transgenic lines in which VND7 activity could be induced post-translationally. This analysis identified 63 putative direct target genes of VND7, which encode a broad range of proteins, such as transcription factors, IRREGULAR XYLEM proteins and proteolytic enzymes, known to be closely associated with xylem vessel formation. more...

Isolation of poplar genes involved in xylem vessel differentiation

(Submitter supplied) Downstream genes of PtVNS genes were explored with inducible expression system using glucocorticoid receptor (GR). Transgenic poplar plants expressing 35S:AtVND7-VP16-GR were treated with dexamethazone (DEX). A number of genes related to the formation of xylem vessels were induced by DEX.

Expression data from Arabidopsis suspension cells overexpressing VND1-GFP

(Submitter supplied) VND1 is a member of NAC-type family transcription factors and has a important role in xylem differentiation. However, how VND1 regulates vascular differentiation is not well known. Here, we identified specific genes downstream of VND1 with transformed suspension culture cells in microarray experiments.

Ectopic expression of MYB46 identifies transcriptional regulatory genes in secondary wall biosynthesis in Arabidopsis

(Submitter supplied) MYB46 functions as a transcriptional switch that turns on the genes necessary for secondary wall biosynthesis. Elucidating the transcriptional regulatory network immediately downstream of MYB46 is crucial to our understanding of the molecular and biochemical processes involved in the biosynthesis and deposition of secondary walls in plants. The transcription factors identified here may include direct activators of secondary wall biosynthesis genes. more...
