Genome-wide analysis of the altered gene expression in the transgenic mice ectopically expressing the human SRY. [lung]

(Submitter supplied) Analysis of the effects of an ectopically expressed human SRY (hSRY) on gene expression in the mouse lung. We hypothesize that aberrant expression of the human SRY gene could affect the development and physiology of transgenic mice. To test this hypothesis we have established a Cre-LoxP transgene activation system, thereby activating and expressing the hSRY transgene at the single-cell embryonic stage. more...

Role of sex chromosome complement in autosomal gene expression

(Submitter supplied) It has generally been assumed that most differences between males and females are due to developmental and hormonal differences between the sexes. Here we investigate the contribution of sex chromosomal complement to such sexual dimorphisms. These genome-wide transcription profiling showed that the expression of hundreds of autosomal genes was sensitive to sex chromosome complement, rather than gender. more...

Gonadal Identity in the Absence of pro-Testis Factor SOX9 and pro-Ovary Factor beta-catenin

(Submitter supplied) The main goal of our study is to identify the molecular events that determine the gonadal identity in mammals. Although testis and ovary arise from a common embryonic primordium, they represent outcomes of opposing fate determination. This decision to differentiate into a testis or an ovary hinges upon the balance between two antagonizing factors, pro-testis SOX9 and pro-ovary β-catenin. This microarray analysis led to the identification of the genes involved in the fate of XX and XY gonads in absence of SOX9 and beta-catenin

Expression data from Mouse embryo

(Submitter supplied) Mice lacking the function of the PcG protein CBX2 (also known as M33) show defects in gonadal, adrenal, and splenic development. In particular, XY knockout mice develop ovaries but not testes, and the gonads are hypoplastic in both sexes. To clarify the molecular basis of the defects, RNA prepared from both wild type and KO gonads were subjected to microarray analyses.

Polycomb group protein CBX2 deficiency effect on XY and XX embryonic gonads

Analysis of E11.5 XY and XX embryonic gonads deficient for the Polycomb group protein CBX2. Animals lacking CBX2 exhibit defects in gonadal, adrenal and splenic development. Results provide insight into role of Cbx2 in gonadal sex differentiation.

Stochastic anomaly of methylome but persistent SRY hypermethylation in disorder of sex development in canine somatic cell nuclear transfer

(Submitter supplied) To infer potential epigenetic variations during SCNT procedure that could occur and lead to abnormal sex-reversal phenotype, we performed whole genome bisulfite sequencing (WGBS) on DNAs extracted from primary fibroblast cells isolated from seven dog samples. These samples include two normal donors, two sex-reversed cloned dogs, one sex-reversed re-cloned dog as well as a pair of independent replicate samples taken as controls
