SOX9 targets in hESC-derived pancreatic progenitors

(Submitter supplied) Our lab identified Sox9 as a specific marker and maintenance factor of mouse pancreatic progenitors (Seymour et al., PNAS, 2007). Here was wanted to identify direct targets of Sox9 in pancreatic progenitors. However, due to the limited number of pancreatic progenitors in the developing mouse, we used in vitro derived pancreatic progenitors to determine direct targets of Sox9. We performed ChIP-seq analysis for Sox9 and determined its direct targets in the human genome. more...

hESC-derived liver transcriptome analysis

(Submitter supplied) To characterize the transcriptional programs that underlie pancreas differentiation and identity, we have generated genome-scale expression profiles by RNA-seq from human embryonic stem cell derived liver progenitors and human fetal pancreatic tissue (days 54-57 post conception). These samples were compared to those already published transcriptomes (Xie et al., 2013). Together, these samples were used to perform principles compotent analysis. more...

Single-cell transcriptomic analyses reveal distinct dorsal/ventral pancreatic programs

(Submitter supplied) The pancreas of vertebrates is separately derived from both the dorsal and ventral endodermal domains. However, the difference between these two programs has been unclear. Here, using a pancreatic determination gene, Pdx1, driven GFP transgenic mouse strain, we identified Pdx1-GFP highly expressing cells (Pdx1high) and Pdx1-GFP lowly expressing cells (Pdx1low) in both embryonic dorsal Pdx1-expressing region (DPR) and ventral Pdx1-expressing region (VPR). more...

In vivo and in vitro Genome wide binding (ChIP-Seq) of Bapx1 and Sox9

(Submitter supplied) Bapx1 was endogenously tagged with S-Peptide and dissected vertebral columns from these tagged embryos at E12.5 were subjected to immunoprecipitation by S-peptide antibody. Simultaneously V5 tagged Bapx1 was also over expressed in NIH_3T3 cells and immunoprecipitated with V5 antibody. Concurrently vertebral column fro E12.5 mouse embryos were subjected to immunoprecipitation with Sox9 antobody to compare binding sites of Sox9 and BApx1 in the vertebral column of developing mouse embryo

SOX9+/PTF1A+ Cells Define the Tip Progenitor Cells of the Human Fetal Pancreas of the Second Trimester

(Submitter supplied) Significant progress has been made in recent years in characterizing human multipotent progenitor cells (hMPCs) of the early pancreas; however, the identity and persistence of these cells during the second trimester, after the initiation of branching morphogenesis, remain elusive. Additionally, studies on hMPCs have been hindered by few isolation methods that allow for the recovery of live cells. Here, we investigated the tip progenitor domain in the branched epithelium of human fetal pancreas between 13.5 and 17.5 gestational weeks by immunohistological staining. more...
