Differential Responses of Plasmacytoid Dendritic Cells to Influenza and Distinct Viral Pathogens

(Submitter supplied) Plasmacytoid dendritic cells (pDCs) are key components of the innate immune response that are capable of synthesizing and rapidly releasing vast amounts of type I interferons (IFNs), particularly IFN-alpha. Here we investigated whether pDCs, often regarded as a mere source of IFN, discriminate between various functionally discrete stimuli and to what extent this reflects differences in pDC responses other than IFN-alpha release. more...

Proteomic and single-cell transcriptomic dissection of human plasmacytoid dendritic cell response to influenza virus

(Submitter supplied) Plasmacytoid dendritic cells (pDCs) represent a rare innate immune subset uniquely endowed with the capacity to produce substantial amounts of type-I interferons (IFN-I). This function of pDCs is critical for effective antiviral defenses and has been implicated in autoimmunity. While IFN-I and select cytokines have been recognized as pDC secreted products, a comprehensive agnostic profiling of the pDC secretome in response to a physiologic stimulus has not been reported. more...

Impact of influenza A on human plasmacytoid dendritic cells (pDC) gene expression

(Submitter supplied) Over 2600 genes were differentially expressed in pDCs exposed to influenza A compared with the controls (no virus). Multiple functional group clusters of genes were impacted, including those involving antiviral responses and also metabolism, such as the glycolysis, oxidative phosphorylation, glucose metabolic process, and positive regulation of macromolecule biosynthesis clusters.

Human plasmacytoid dendritic cells mount a distinct antiviral response to virus-infected cells

(Submitter supplied) Plasmacytoid dendritic cells (pDCs) can rapidly produce interferons and other soluble factors in response to extracellular viruses or virus mimics such as CpG-containing DNA. pDCs can also recognize live cells infected with certain RNA viruses, but the relevance and functional consequences of such recognition remain unclear. We studied the response of primary DCs to the prototypical persistent DNA virus, the human cytomegalovirus (CMV). more...

Eosinophil-mediated suppression and Anti-IL-5 enhancement of plasmacytoid dendritic cell interferon responses in asthma

(Submitter supplied) Blood plasmacytoid dendritic cells (pDCs) purified from anonymous donors were stimulated ex vivo with rhinovirus-16 (RV) in the presence or absence of eosinophil supernatants. Purified pDCs from 8 individuals with moderate-severe asthma, treated or not treated with anti-IL-5/5Rα therapy, were cultured with or without RV. Transcriptome analysis revealed global repression of pDC IFN response patterns by eosinophils, most notably in basal expression of interferon stimulated genes (ISGs). more...

Response of IRF7-deficient peripheral blood mononuclear cells to pH1N1 influenza virus infection

(Submitter supplied) IRF7 plays a critical role in the production and amplification of the antiviral type I and III interferon response. Autosomal recessive IRF7-deficiency resulted in life-threatening influenza disease in a 3-year-old child. We studied the impact of IRF7-deficiency in non-hematopoietic tissues (fibroblasts and lung epithelial cells) as well in hematopoietic cells (peripheral blood mononuclear cells (PBMCs)). more...

Single cell RNA sequencing-based comparison of mouse splenic pDC, pDC-like cells, tDC, cDC1 and cDC2 at steady state and during a viral infection

(Submitter supplied) Plasmacytoid dendritic cells (pDC) are the main source of type I interferon (IFN) during viral infections. Their other functions are debated, due to a lack of tools to identify and target them in vivo without affecting pDC-like cells and transitional DC (tDC), which harbor overlapping phenotypes and transcriptomes but a higher efficacy for T cell activation. To overcome this bottleneck, we designed, generated and validated a pDC-Tomato reporter mouse. more...

Single cell transcriptomic atlas of human plasmacytoid dendritic cells reveals a distinct cytotoxic-like subset in HIV-1 infected individuals.

(Submitter supplied) To understand how the transcriptome of pDCs is impacted by HIV-1 infection and exogenous stimulation, we sorted pDCs from people with HIV-1 (PWH) on antiretroviral treatment before and after toll-like receptor 9 (TLR9) agonist treatment, as well as from healthy controls, and performed single-cell (sc)-RNA sequencing. We generated a transcriptomic atlas of over 112,000 pDCs. Our cluster analysis revealed an inducible cytotoxic-like pDC cluster characterized by high expression of both antiviral and cytotoxic genes. more...

Single-cell RNA-sequencing reveals pervasive but highly cell type-specific genetic ancestry effects on the response to viral infection

(Submitter supplied) Humans show remarkable variation in susceptibility to infectious diseases as well as chronic inflammatory and autoimmune disorders. This heterogeneity arises partially from variation in the immune response, which is responsible for preventing and controlling infection. To better understand the major factors driving antiviral immune response differences, we used single-cell RNA-sequencing to measure the effects of genetic ancestry and cis-regulatory variation on the transcriptional response to influenza infection in various immune cell types in 90 European and African American individuals. more...

Gene expression analysis of hPSC-derived macrophages (iMACs) encountering Streptococcus pneumoniae.

(Submitter supplied) Purpose: to analyze gene expression changes of iMACs encountering S. pneumoniae. Methods: SMART-seq2 amplified polyA RNA from sorted iMACs in the co-cultures. Results: the transcriptome of iMACs changed after encountering S. pneumoniae.
