Stem cell factor programs the mast cell activation phenotype

(Submitter supplied) Mast cells, activated by antigen via the high affinity receptor for IgE (FcεRI), release an array of pro-inflammatory mediators that contribute to allergic disorders such as asthma and anaphylaxis. The KIT ligand, stem cell factor (SCF), is critical for mast cell expansion, differentiation and survival, and, under acute conditions, enhances mast cell activation. Stem cell factor effect on bone marrow-derived mast cells

Analysis of bone marrow-derived mast cells (MC) treated with interleukin 3 (IL3) or IL3 + stem cell factor (SCF). MCs chronically exposed to SCF have a greatly reduced capacity to degranulate and to generate cytokines in response to antigen. Results provide insight into role of SCF in MC activation.

Murine bone marrow-derived mast cells (BMMCs) transduced with pAPM lentiviruses overexpressing miR-221 vs control overexpressing miR-221m.

(Submitter supplied) Bone marrow-derived mast cells were derived from three different female C57Bl/6 mice of similar age. Cells were transduced with pAPM lentiviruses either overexpressing microRNA-221 or a non functional microRNA-221 mutant, termed miR-221m, where four point-mutations were introduced in the seed sequence of miR-221.
