Transcriptome analysis of hpl-2(tm1489) vs wild-type worms at mixed embryonic and third larval stages

(Submitter supplied) Epigenetic factors guide cell fate decisions and can stabilize development by buffering environmental variation. HP1 proteins have a well-characterized role in heterochromatin packaging and gene regulation, while their function in organismal development is less well understood. Here we used genome-wide expression profiling to assess novel functions of the C. elegans HP1 homologue HPL-2 at specific developmental stages

Regulation of signaling genes by TGFbeta during entry into dauer diapause in C. elegans

(Submitter supplied) BACKGROUND: When resources are scant, C. elegans larvae arrest as long-lived dauers under the control of insulin/IGF- and TGFbeta-related signaling pathways. : We have identified genes that show different levels of expression in a comparison of wild-type L2 or L3 larvae (non-dauer) to TGFbeta mutants at similar developmental stages undergoing dauer formation. Many insulin/IGF pathway and other known dauer regulatory genes have changes in expression that suggest strong positive feedback by the TGFbeta pathway. more...

Tissue-specific ChIP-seq of C. elegans Rb/E2F

(Submitter supplied) The Rb/E2F tumor suppressor regulates gene expression to control the onset and timing of differentiation in many different cell types during development. This critical function makes Rb/E2F a frequent target for inactivation in tumors of diverse tissue origin. However, the mechanisms by which Rb/E2F governs tissue-specific gene regulation in vivo are poorly understood. We have determined the genome-wide binding profiles for components of the C. more...

Transcriptional repression of Hox genes by binding of HP1 and methylated H1 to H3K27me3 in C. elegans

(Submitter supplied) Elucidation of the biological role of H1 and HP1 in mammals has been difficult owing to the existence of a least 11 distinct H1 and three HP1 subtypes in mice. Therefore we decided to study the function of HP1 (HPL-1 and HPL-2) and H1 (HIS-24) related proteins in C. elegans, since hpls and his-24 deficient nematodes are viable. Global transcriptional analysis of the mutant animals revealed that expression of only a small number of genes was affected and HPLs, and HIS-24 appear to influence the expression of genes through either activation or repression. more...

The C. elegans ortholog of TDP-43 regulates the chromatin localization of the heterochromatin protein 1 homolog, HPL-2

(Submitter supplied) TDP-1 is the C. elegans ortholog of mammalian TDP-43, which is strongly implicated in the etiology of Frontotemporal Dementia (FTD) and Amyotrophic Lateral Sclerosis (ALS). We discovered that deletion of the tdp-1 gene results in enhanced transcriptional gene silencing leading to increased sensitivity to heritable RNA interference (RNAi). As heritable RNAi in C. elegans depends on chromatin changes moderated by HPL-2, a homolog of heterochromatin protein 1 (HP1), we investigated the interaction of TDP-1 and HPL-2. more...

sRNA profiles of control adults, postdauer adults, larva L3, and dauer stages of C. elegans

(Submitter supplied) Early environmental experiences and life histories profoundly influence adult phenotypes via as yet poorly understood mechanisms. We previously showed that wild-type adult C. elegans that transiently passed through the stress-induced dauer larval stage (post-dauers) exhibit different gene expression patterns, genome-wide chromatin structure, and life history traits when compared to adults that bypassed the dauer stage (controls). more...

Novel roles of C. elegans heterochromatin protein HP1 and linker histone HIS-24 in the regulation of innate immune gene expression and stress Response

(Submitter supplied) Linker histone H1 and heterochromatin protein 1 (HP1) are essential components ofheterochromatin which contribute to the transcriptional repression of genes. It has been shown that the methylation mark on the histone H1 serves as a specific recognition code for the chromodomain of HP1, however, the functional role of the HP1/H1 complex remains elusive. Using C. elegans, we elucidate the function of the linker histone variant HIS-24 and heterochromatin proteins HPL/HP1 in the cooperative transcriptional regulation of immunerelevant and stress resistance genes. more...

Gene expression C. elegans age-1 mutants with neuron- or intestine-restricted age-1 activity

(Submitter supplied) The goal of this experiment was to identify transcripts regulated non-autonomously by age-1 activity in neurons or intestine. Comparisons of gene expression were made between age-1(mg44) null adults and CY251 (age-1(mg44); bvIs2) and CY262 (age-1(mg44); bvIs1); all were relative to wildtype (N2). The integrated transgenes, bvIs2 and bvIs1, carry the age-1 cDNA expressed from the neuronal ric-19 promoter (bvIs2) or intestinal ges-1 promoter (bvIs1). more...

daf-16/FoxO isoform-specific deletion mutants

(Submitter supplied) FoxO transcription factors promote longevity across taxa. How they do so is poorly understood. In the nematode Caenorhabditis elegans, the A- and F-isoforms of the FoxO transcription factor DAF-16 extend life span in the context of reduced DAF-2 insulin-like growth factor receptor (IGFR) signaling. To elucidate the mechanistic basis for DAF-16/FoxO-dependent life span extension, we performed an integrative analysis of isoform-specific daf-16/FoxO mutants. more...
