Expression data 550cGy vs 0cGy KSL

(Submitter supplied) Precise study of HSCs during regeneration has been impeded by the rarity of the HSC population and depletion of phenotypic HSCs early following genotoxic stresses, such as total body irradiation (TBI). We isolated bone marrow (BM) ckit+sca-1+lin- (KSL) cells, which are enriched for HSCs, from adult C57Bl6 mice before and at several time points following TBI, as a means to map the dynamic molecular response of HSC regeneration.

The induction of integrin beta3 signaling in hematopoietic stem cells (HSCs)

(Submitter supplied) To determine the molecules involved in beta3 integrin signaling in HSCs, we carried out DNA microarray analyses using HSCs stimulated with ligand-mimic antibody for intetegrin beta3 in the presence of stem cell factor(SCF) and/or thrombopoietin (TPO). All experiments were performed in duplicate or triplicate using sorted cells that were independently prepared in separate experiments.

RNAseq analysis of murine ITPKB deficient versus wild type LT-HSC

(Submitter supplied) Tight regulation of hematopoietic stem cell (HSC) homeostasis is essential for life-long hematopoiesis, for preventing blood cancers and for averting bone marrow failure. The underlying mechanisms are incompletely understood. Here, we identify production of inositol-tetrakisphosphate (IP4) by inositoltrisphosphate 3-kinase B (ItpkB) as essential for HSC quiescence and function. Young ItpkB-/- mice accumulated phenotypic HSC and showed extramedullary hematopoiesis. more...

Gene expression analysis of hematopoietic stem cells based on Syndecan-2 surface expression

(Submitter supplied) Analysis of how Syndecan-2 expression identifies hematopoietic stem cells with unique gene expression profiles. We tested the hypothesis that Syndecan-2 expression enriches for hematopoietic stem cell gene expression programs. The results provide important insight into how Syndecan-2 can be used to isolate hematopoietic stem cells with distinct hematopoietic properties.

Aryl hydrocarbon receptor antagonists promote the expansion of human hematopoietic stem cells

(Submitter supplied) Although practiced clinically for more than 40 years, the use of hematopoietic stem cell (HSC) transplants remains limited by the ability to expand these cells ex vivo. An unbiased screen with primary human HSCs identified a purine derivative, StemRegenin 1 (SR1), that promotes the ex vivo expansion of CD34+ cells. Culture of HSCs with SR1 led to a 50-fold increase in cells expressing CD34 and a 17-fold increase in cells that retain the ability to engraft immunodeficient mice. more...

StemRegenin 1 effect on hematopoietic stem cells: dose response

Analysis of hematopoietic stem cells (HSC) treated with StemRegenin 1 (SR1) and LGC006 (a less potent SR1 analog) at 30nM to 1000nM. The use of HSC transplants depends on the ability to expand these cells ex vivo. Results provide insight into molecular basis of SR1-induced ex vivo HSC expansion.

Transcriptome data of bone marrow hematopoietic stem cells (HSCs) with IGF-1 treatment

(Submitter supplied) Insulin-like growth factor-1 (IGF-1) is known as a hematopoietic factor which impacts hematopoietic reconstitution and facilitates thrombopoiesis, although its direct effect on hematopoietic stem cells (HSCs) remains unclear. Here, we show that IGF-1 rapidly prompts megakaryocyte (Mk)-lineage differentiation of HSCs by enhancing mitochondrial activity in HSCs. To identify the phenotype switching and function variation in HSCs with IGF-1 treatment, we performed RNA-seq of HSCs from the bone marrow of mice with single-dose IGF-1 treatment (IGF-1) and their age-matched control mice (Ctrl).

Gene expression signatures of megakaryocytes post-IR

(Submitter supplied) Platelets are anucleated blood cells that are produced by their progenitor cell, the megakaryocyte (MK). Platelets are centrally positioned in hemostasis and thrombosis and, according to the recent findings, play key roles in innate immunity, inflammation, atherogenesis, and cancer metastasis. The quantitative and qualitative properties of platelets are crucial determinants of their hemostatic function. more...

Transcriptomic analysis of Brm-/- hematopoietic stem and progenitor cells (LSK)

(Submitter supplied) Little is known of hematopoietic stem (HSC) and progenitor (HPC) cell self-renewal. The role of Brahma (BRM), a chromatin remodeler, in HSC/HPC function is unknown. Bone marrow (BM) from Brm-/- mice manifested increased numbers of long- and short-term HSCs, GMPs, and increased numbers and cycling of functional HPCs. Brm-/- HSC/HPC show demonstrated functional differences compared to wildtype HSC/HPC in vivo and ex vivo, due in part to increased intracellular valine in lineage negative BM. more...

Single cell transcriptome sequencing of expanded human hematopoieic stem/progenitor cells cultured in PCL-PVAc-PEG based 3a medium and StemSpan with UM171/SR-1 medium.

(Submitter supplied) We demonstrated that chemically difined PCL-PVAc-PEG based 3a medium was superior to conventional StemSpan besed medium in expansion of human hematopoietic stem cells. In fact, more phenotypic HSCs (CD34+EPCR+CD90+CD45RA-ITGA3+) were expanded in our culture condition and higher donor chmerism was achieved in transplantation assay using humanized mice. But detail transcriptome profiles of expanded HSCs was unknown and therefore single-cell RNA sequencing was done to reveal it. more...

Transcriptomes of cultured human hematopoietic stem cells.

(Submitter supplied) We compared transcriptomes of CD34(high)EPCR-positive with CD34(high)EPCR-negative cells after 10-days culture in PCL-PVAc-PEG based 3a medium. It was because our culture condition had not previously reported and so we demonstrated that phenotypic HSCs were concentrated in EPCR-positive fraction as conventional UM171-based culture system. As a result, CD34(high)EPCR-positive cells highly expressed HSC markers (HLF, AVP, PRDM16 and FGD5) compared with CD34(high)EPCR-negative cells.

Hematopoietic stem cells from mice treated with G-CSF or saline alone for 7 days

(Submitter supplied) Recent studies demonstratethat inflammatory signals regulate hematopoietic stem cells (HSCs). Granulocyte-colony stimulating factor (G-CSF) is often induced with infection and plays a key role in the stress granulopoiesis response. However, its effects on HSCs are less clear. Herein, we show that treatment with G-CSF induces expansion and increased quiescence of phenotypic HSCs, but causes a marked, cell-autonomous HSC repopulating defect associated with induction of toll-like receptor (TLR) expression and signaling. more...

Expression data from WT, p110α-/-, p110δ-/-, and p110αδ double knockout HSC and LMPP hematopoietic populations

(Submitter supplied) Many cytokines and chemokines that are important in hematopoiesis activate the phosphatidylinositol 3-kinase kinase(PI3K) signaling pathway. Because this pathway is frequently mutated and activated in cancer, PI3K inhibitors have been developed for the treatment of several malignancies, and are now being tested in the clinic in combination with chemotherapy. However, the role of PI3K in adult hematopoietic stem cells (HSCs), particularly during hematopoietic stress, is still unclear. more...

Attenuation of miR-126 activity expands HSC in vivo without exhaustion

(Submitter supplied) Life-long blood cell production is governed through the poorly understood integration of cell-intrinsic and -extrinsic control of hematopoietic stem cell (HSC) quiescence and activation. MicroRNAs (miRNAs) coordinately regulate multiple targets within signaling networks making them attractive candidate HSC regulators. We report that miR-126, a miRNA expressed in HSC and early progenitors, plays a pivotal role in restraining cell cycle progression of HSC in vitro and in vivo. more...

RNA-seq in human hematopoietic stem cells (HSCs) cultures on 3 different materials

(Submitter supplied) Hematopoietic stem cell transplantation (HSCT) is successfully applied since the late 1950s, however, its efficacy still needs to be improved. A promising strategy is to transplant high numbers of pluripotent hematopoietic stem cells (HSCs). Therefore, an advanced ex vivo culture system is needed that supports the proliferation and maintains the pluripotency of HSC to override possible limitations in cell numbers gained from donors. more...
