Dysregulated gene expression networks in human acute myelogenous leukemia stem cells

(Submitter supplied) We performed the first genome-wide expression analysis directly comparing the expression profile of highly enriched normal human hematopoietic stem cells (HSC) and leukemic stem cells (LSC) from patients with acute myeloid leukemia (AML). Comparing the expression signature of normal HSC to that of LSC, we identified 3,005 differentially expressed genes. Using 2 independent analyses, we identified multiple pathways that are aberrantly regulated in leukemic stem cells compared with normal HSC. more...

Gene expression profiles of AML derived stem cells: similarity to hematopoietic stem cells

(Submitter supplied) Tumors contain a fraction of cancer stem cells that maintain the propagation of the disease. The CD34‏CD38_ cells, isolated from acute myeloid leukemia (AML), were shown to be enriched leukemic stem cells (LSC). We isolated the CD34‏CD38_ cell fraction from AML and compared their gene expression profiles to the CD34‏CD38‏ cell fraction, using microarrays. We found 409 genes that were at least twofold over- or underexpressed between the two cell populations. more...

Acute myeloid leukemia CD34+CD38- cell population

Analysis of CD34+CD38- and CD34+CD38+ cell fractions isolated from 5 acute myeloid leukemia (AML) patients. AML CD34+CD38- cells are enriched leukemic stem cells (LSC) that maintain the propagation of the disease. Results provide insight into molecular mechanisms that maintain and characterize LSC.

Comparison study of glioma stem cells and adult human neural stem cells

(Submitter supplied) Glioblastoma, the most common and malignant brain tumor, harbors stem-like cells that self-renew and propagate upon serial transplantation. Although they share functional, morphological and developmental similarities to adult brain neural stem cells, stem cell characteristic pathways contributing to glioblastoma stem-like cells have not been consistently determined. Towards this goal we have provided an internally coherent molecular reference that compares adult neural and glioblastoma stem cells cultivated under identical conditions. more...

Next Generation Sequencing of LSK EpCAM+ and EpCAM- of Zeb1-/-

(Submitter supplied) Zeb1, a zinc finger E-box binding homeobox epithelial-mesenchymal (EMT) transcription factor, confers properties of ‘stemness’, such as self-renewal, in cancer. Yet little is known about the function of Zeb1 in adult stem cells. Here, we used the hematopoietic system, as a well-established paradigm of stem cell biology, to evaluate Zeb1 mediated regulation of adult stem cells. We employed a conditional genetic approach using the Mx1-Cre system to specifically knockout (KO) Zeb1 in adult hematopoietic stem cells (HSCs) and their downstream progeny. more...

Gene expression data from sorted primary human cord blood samples

(Submitter supplied) Experiments using xenografts show that some solid tumours and leukemias are organized as cellular hierarchies sustained by cancer stem cells (CSC). Despite promise, the relevance of the CSC model to human disease remains uncertain. Here we show that acute myeloid leukemia (AML) follows a CSC model based on sorting multiple populations from each of 16 primary human AML samples and identifying which contain leukemia stem cells (LSC) using a sensitive xenograft assay. more...

Expression data from chronic myeloid leukemia and normal bone marrow stem and progenitor cells

(Submitter supplied) Chronic myeloid leukemia is a disease originated at the level of hematopoietic stem cell, characterized by the abnormal overproduction and accumulation, both in blood and bone marrow, of myeloid cells. Treatment options include tyrosine kinase inhibitors that inhibit BCR-ABL activity, however some patients develop resistance to these drugs and has been asociated to the stem cells We have performed a comparative analysis of the global gene expression profiles between CML and normal HSCs. more...

Expression profiling of murine hemopoietic cell survival on cDNA arrays.

(Submitter supplied) Deregulated cell survival programs are a classical hallmark of cancer. We have previously identified a serine residue (Ser585) in the beta-c subunit of the granulocyte-macrophage colony-stimulating factor (GM-CSF) receptor that selectively and independently promotes cell survival. We now show that Ser585 phosphorylation is constitutive in 20/23 (87%) acute myeloid leukemia (AML) patient samples indicating that this survival-only pathway is frequently deregulated in leukemia. more...

Expression data from human hematopoietic stem and progenitor compartments from patients with acute myeloid leukemia with monosomy 7 and healthy controls

(Submitter supplied) We applied a novel approach of parallel transcriptional analysis of multiple, highly fractionated stem and progenitor populations in a genetically defined subset of AML (AML with monosomy 7). We isolated phenotypic long-term HSC (LT-HSC), short-term HSC (ST-HSC), and committed granulocyte-monocyte progenitors (GMP) from individual patients with AML, and measured gene expression profiles of each population, and in comparison to their phenotypic counterparts from age-matched healthy controls.

Activation or maintenance of a leukemia stem cell self-renewal pathway in downstream myeloid cells

(Submitter supplied) Activation or maintenance of a leukemia stem cell self-renewal pathway in downstream myeloid cells is an important component of AML development We generated either MLL-AF9 mediated murine leukemias that originate from committed progenitor (GMP) cells or Hoxa9/Meis1a mediated murine leukemias that originate from hematopoietic stem cells (HSC). The leukemia stem cell fraction in these two type of leukemias shared a common self-renewal pathway with normal hematopoietic stem cells. more...

Acute myelogenous leukemia stem cells

Analysis of acute myelogenous leukemia (AML) leukemia stem cells (LSC) induced either by Hoxa9/Meis1a oncogenes or MLL-AF9 oncoprotein. LAA expression profiles on LSC compared to other tissues

(Submitter supplied) Leukemic stem cells (LSC) might be the source for leukemic disease self-renewal and account for disease relapse after treatment, which makes them a critical target for further therapeutic options. Leukemia associated antigens (LAA) might be suitable structures to be attacked by immunotherapeutic agents. We performed primary AML sample enrichment and microarray studies to define LAA expression levels in AML. more...

Gene expression data of human chord blood hematopoietic progenior cells subjected to myeloid differentiation and transduced with lncRNA HOXA10-AS

(Submitter supplied) Long non-coding RNAs (lncRNAs) have emerged as crucial regulators of gene expression and cell fate decisions. We predicted HOXA10-AS as a novel non-coding regulator of hematopoiesis and leukemogenesis We overexpressed HOXA10-AS using a bidirectional vector construct, which resulted in impaired monocytic in vitro differentiation and myeloid colony-formation. In addition, shRNA, sgRNA and LNA-GapmeR mediated downregulation of HOXA10-AS resulted in reduced leukemic growth of KMT2A-r cell lines and patient blasts. more...

Bortezomib suppresses self-renewal and leukemogenesis of leukemia stem cell by NF-ĸB-dependent inhibition of cyclin dependent kinase 6 in MLL-rearranged myeloid leukemia

(Submitter supplied) Acute myeloid leukemia (AML) with chromosomal rearrangements involving the H3K4 methyltransferase mixed-lineage leukemia (MLL) is an aggressive subtype with low overall survival. MLL rearrangements rapidly transform hematological stem and progenitor cell (HSPC) to leukemia stem cell (LSC). Bortezomib (Velcade) is used widely in hematological malignancies. However, it is still unknown whether bortezomib possesses anti-self-renewal and anti-leukemogenesis of LSC in AML with MLL rearrangements. more...
