Diurnal and Circadian-Regulated Genes in Arabidopsis

(Submitter supplied) Plants respond to day/night cycling in a number of physiological ways. At the mRNA level, the expression of some genes changes during the 24-hr period. To identify novel genes regulated in this way, we used microarrays containing 11,521 Arabidopsis expressed sequence tags, representing an estimated 7800 unique genes, to determine gene expression levels at 6-hr intervals throughout the day. Genome-wide identification of CCA1-bound genes in Arabidopsis by ChIP-Seq

(Submitter supplied) LHY and CCA1 encode single MYB transcription factors, involved in circadian clock. However, direct target genes of LHY and CCA1 in a genomic scale were largely unknown. To reveal bound genes by CCA1, chimeric protein CCA1-FLAG was expressed under CCA1 promoter in cca1 lhy (CCA1pro:CCA1-FLAG/ cca1 lhy). ChIP was performed using anti-FLAG antibody (F3165; SIGMA), which was bound to Dynabeads Protein G (100-03D; Life Technologies), and ChIP DNA were analyzed by IonPGM or Illumina GAII.

Diurnal gene expression in Arabidopsis thaliana Col-0 rosette leaves

(Submitter supplied) How do the transcript levels of leaf-expressed genes change in a normal day-night cycle? The interest is in genes that are regulated by the circadian clock and the diurnal component (i.e. light, metabolite changes). Plants were grown on soil in a 12/12 h light/dark rythm at 20°C day and night. 5 weeks after germination the rosettes of the non-flowering plants were harvested, 15 plants per sample. Diurnal cycle effect in leaves: time course

Analysis of Col-0 leaves at various time points in a normal day-night cycle. Col-0 plants grown in 12-hour light 12-hour dark rhythm at 20 degrees C. Leaves collected 4, 8, and 12 h into the light period and into the night. Results identify genes that undergo diurnal changes of expression.
