Design: The construction of SMRTbell libraries suitable for generating high accuracy long reads on the Sequel Systems requires 15 g of high molecular weight genomic DNA (gDNA). The DNA is sheared to a mode of 15 kb using Diagenodes Megaruptor. SMRTbell libraries are prepared from sheared gDNA using PacBios SMRTbell Template Prep Kit 1.0. SMRTbell libraries are then size-fractionated using Sage Sciences SageELF. The fractions suitable for sequencing have insert sizesfrom 10 kb to 15 kb.
