1 ILLUMINA (Illumina HiSeq 2000) run: 2.4M spots, 715.4M bases, 246.7Mb downloads

Design: Genomic DNA of all 50 longan accessions was isolated from 100 mg of young leaf tissue using DNeasy Plant Mini Kit (Qiagen Sciences, USA) according to the manufacturer's instructions. Then, the isolated DNA of each sample was quantified using Nanodrop 8000 spectrophotometer (Thermo Fisher Scientific, USA) and adjusted to the final concentration of 50 ng/ul. Genomic DNA (50 ul) was digested with the type II restriction endonuclease ApeKI to construct GBS libraries according to the protocol described in Elshire et al. (2011). The library was sequenced on an Illumina HiSeq 2000 platform at Novogene Co., Ltd (Hong Kong).

A total of 50 longan accessions from the Longan Community BioBank, Nong Chang Khuen, Lamphun, Thailand, were used in this study. Of these, 48 accessions are Thai cultivars, and the other two cultivars, Chuliang and Kohala, are foreign cultivars. Genomic DNA of all 50 longan accessions was isolated from 100 mg of young leaf tissue using DNeasy Plant Mini Kit (Qiagen Sciences, USA) according to the manufacturer's instructions. Then, the isolated DNA of each sample was quantified using Nanodrop 8000 spectrophotometer (Thermo Fisher Scientific, USA) and adjusted to the final concentration of 50 ng/ul.
