Extraction protocol Tumors were dissected and collected in cold PBS. Tumors were minced and placed in dissociation media consisting of RPMI supplemented with 2 mg/ml collagenase I, 5% FBS and 15 U/ml DNAse. Samples were dissociated using the gentleMACS dissociator (Miltenyi Biotec), and filtered through a 70 μm cell strainer. After red blood cell lysis, cell suspensions were washed with PBS with 0.04% BSA and 6000 cells were loaded into a Single Cell A Chip.
Library preparation was done following manufacturer's instructions for the ChromiumTM Single Cell Reagent Kit v2
